چکیده
Lipolytic enzymes from endophytic microbes have been the focus of intense and growing research in view applicability such various industrial applications microorganisms may emerge as a potential source lipases with distinct characteristics. The current work involved purification characterization lipase an endophytic-bacteria isolated stem Withania somnifera L. Dunal. Based on analysis morphology 16S rDNA sequence, bacteria was identified Bacillus pumilus designated B. WSS5. Purified WSS5 found to be 27KDa revealed by SDS-PAGE optimum activity at 37 °C pH 8. stability enzyme presence metal ions, surfactants, inhibitors, organic solvents suggested that this could exploited leather detergent industries. In addition, stain removal crude purified determined cotton fabric pieces stained vegetable oil. Lipase added water along significantly enhanced efficiency commercially available detergent. findings suggest pumilusWSS5 owing promising properties alkali-thermostability, solvent-tolerance, broad substrate specificity makes it strong candidate for application formulations, biotransformation, industries, medicine. To our knowledge, is first report bacterial endophyte

Lipase from Bacillus pumilus RK31: Production, Purification and Some Properties
A new lipolytic strain of Bacillus sp. was isolated from oil contaminated soil sample. On measurement of biochemical parameters and 16S rDNA sequence information the strain was identified as Bacillus pumilus RK31 (NCBI accession no GQ463238). The lipase purification steps involved, 60% ammonium sulphate saturation, gel filtration chromatography using Sephadex G200 and ion exchange chromatograph...
متن کاملMolecular Cloning and Characterization of a Lipase from an Indigenous Bacillus pumilus
Cloning and sequencing of a lipase gene from an indigenous Bacillus pumilus, strain F3, revealed an open-reading frame of 648 nucleotides predicted to encode a protein of 215 residues. Sequence analysis showed that F3 lipase contained a signal peptide composed of 34 amino acids with an H domain of 18 residues. A tat-like motif was found in the signal peptide similar to some other Bacillus pumil...
متن کاملPurification and Characterization of Organic Solvent and Detergent Tolerant Lipase from Thermotolerant Bacillus sp. RN2
The aim of this study was to characterize the organic solvent and detergent tolerant properties of recombinant lipase isolated from thermotolerant Bacillus sp. RN2 (Lip-SBRN2). The isolation of the lipase-coding gene was achieved by the use of inverse and direct PCR. The complete DNA sequencing of the gene revealed that the lip-SBRN2 gene contains 576 nucleotides which corresponded to 192 deduc...
متن کاملPurification and Characterization of a Fibrinolytic Enzyme from Bacillus pumilus 2.g Isolated from Gembus, an Indonesian Fermented Food
Bacillus pumilus 2.g isolated from gembus, an Indonesian fermented soybean cake, secretes several proteases that have strong fibrinolytic activities. A fibrinolytic enzyme with an apparent molecular weight of 20 kDa was purified from the culture supernatant of B. pumilus 2.g by sequential application of ammonium sulfate precipitation, ion-exchange chromatography, and hydrophobic chromatography....
